ABSTRACT

Objective:To explore the efficacy and safety of improved skin expanding method on patients with arm infusion port. Methods:Clinical data of 100 patients with gynecological malignant tumor receiving adjuvant chemotherapy was retrospectively analyzed, including 50 patients treated by traditional way of expanding skin(transverse and micro intubation sheath inserted into blood vessels) and 50 patients treated by modified skin expanding method, namely making an incision with both depth and length of 0.5 cm which tilting 45° from the thread toward the port. The indexes as planting time, smooth operation, incidence of secondary enlargement, patient satisfaction, port moving time and incidence of postoperative complications were compared and analyzed between the two groups.Result:Modified skin expanding method was superior to traditional skin expanding way in planting time, smooth operation, incidence of secondary enlargement, patient satisfaction, port moving time and incidence of postoperative complications. Conclusions:Compared with the traditional way of expanding skin, modified skin expanding method can adjust catheter malposition caused by connecting with the port and send the catheter back into blood vessel at the incision maintaining the smooth of the catheter. Can make the operation well going increasing the efficiency of implantation of the upper arm infusion port. Can avoid the adverse consequences from the straddle of blood vessel by catheter and port in the tunnel after the catheter indwelling, which greatly enhances the safety of patients during infusion port application period.

ABSTRACT

Objective To construct a recombinant plasmid vector containing the distal fragment of the distal C-terminus (dDCT) of the Cav 1.2 channel, and express, extract, and purify dDCT protein and characterize its biological activity. Methods dDCT cDNA was ligated into the pGEX-6p-1 vector to create a recombinant plasmid that was subsequently transformed into Escherichia coli BL21 competent cells.Expression of GST-dDCT fusion protein from this plasmid was induced with isopropy-β-D-thiogalactoside,and the resulting protein was purified using glutathione-sepharose 4B beads. The biological activity of dDCT was analyzed by GST pull-down assay. Results The recombinant plasmid was verified by restriction enzyme digestion and sequencing. The concentration and purity of the dDCT protein, which was extracted by ultrasonication, were high enough to detect dDCT activity. The binding of dDCT to CT1 was determined to be concentration-dependent.Conclusion The recombinant dDCT plasmid was successfully constructed, providing the fundamental basis for future studies on mechanisms of Cav 1.2 channel autoregulation.

ABSTRACT

Objective To evaluate the characteristics of the PREVI Isola automated plate streaker (bioMérieux,SA).Methods 80 respiratory tract specimens,70 sterile fluids,52 stools,69 swabs,12 cerebrospinal fluids and 80 urines were collected in Peking Union Medicd College Hospital.Specimens were processed with manual streaking and PREVI Isola system.PREVI Isola system were evaluated comparing to the manual streaking. The quality of results were analyzed by SPSS 16.0, doing Wilcoxon’s Sign Rank Test for the results of finally isolated species, overall numbers of isolated colonies and semi-quantita-tive of the species were both isolated by the two methods. Results PREVI Isola system was highly automatic, which could select the right plates and stake the bar code on the back of the plate indicating the type of the agar and inoculation time, PREVI Isola system could accurately absorb the liquid specimen and use a novel comb streaking procedure for processing of fluid specimens on standard agar plates, like 17 inoculating loops work together. It also had a good reproducibility. The quali-ty of PREVI Isola system results:As to the finally isolated species, there was significant statistical difference between PREVI Isola system and manual streaking method in respiratory tract and stools specimen, there were more species isolated by manual streaking method than PREVI Isola system. There were no differences between the two methods for the other of specimen types. As to the amount of pure clones of the species were both isolated by the two methods, there were significant statistical differences between the two methods for respiratory tract, sterile fluid and stool specimens. The amount of clones isolated by PREVI Isola system was more than manual streaking method. In semi-quantitative results, there were significant statistical differences between the two methods for respiratory tract and urine specimen,Species had wider distribution of PREVI Isola system than manual streaking method.Inoculation efficiency:if the batch of specimen type was simple (mainly the urine and so on),using the same plates,PREVI Isola system was more efficient than manual streaking method.Howev-er,if the batch of specimen type was complicated, manual method was high-performance.Besides,not all specimen type could be inoculated by PREVI Isola system, such as cerebrospinal fluid, catheter and tissues. Conclusion If the lab had simple specimen type,or utilize the specimen type using the same agar plates to be inoculated together,PREVI Isola system belongs to a good performance automated plate streaker.

ABSTRACT

The rearrangement reaction of benzyl phenyl ethers (BPE) plays an important role in the organic chemistry and drug synthesis .This paper briefly reviewed the rearrangement reaction and its possible mechanism in various catalytic sys-tems ,such as trifluoroacetic acid ,phosphotungstic acid ,cyclodextrin ,molten tin and aluminium bromide .
